Neurogenesis occurs in the adult mammalian brain in two specialized geminal zones: the subventricular zone (produces new neurons destinated for the olfactory bulb) and the granular cell layer of the dentate gyrus of the hippocampus.
We examine of the effects of neuroprotective, neuroregenerative and neuromodulatory compounds on cell composition, cell proliferation, neurogenesis and protein expression after ischemic insults.
The mitotic marker BrdU (Bromodeoxyuridine) is used as marker for cell proliferation in both models. Neurogenesis and gliogenesis are characterised by cell specific marker (NeuN, Doublecortin and S100-ß, GFAP, OX42 for example). Analysis of fluorescence double labelling is performed by confocal laser microscopy.
