To compare confocal laserscanning microscopy (top) and two photon laserscanning microscopy (bottom) with respect to resolution, penetration and bleaching 3D-image data sets from Lucifer Yellow injected neurons were taken by exciting the tissue first with ultrashort laser pulses (puls width: 1,3 ps, wavelength: 800 nm) and subsequently with a continuous laser beam (wavelength: 476/488 nm). The images demonstrate that dendritic spines are clearly visible with both excitation methods indicating that image quality of 2P LSM compares well with CLSM. Isosurface volume rendering was performed using a 3D image sComparison of 1Photon- versus 2 Photon-excitationoftware (AMIRA, Indeed, Berlin).
