Forschungsgruppe Neuroplastizität

Schwerpunktthemen unserer Forschergruppe sind zelluläre und molekulare Mechanismen neuronaler Plastizität im gesunden Gehirn, aber auch Störungen synaptischer Funktionen bei neuropsychiatrischen und neurodegenerativen Erkrankungen – so genannte Synaptopathien. Wir untersuchen die molekulare Dynamik von Postsynapsen, die Entschlüsselung von neuronalen Calcium-Signalen durch Calcium-Sensor-Proteine, die Kommunikation von Synapsen mit dem Zellkern und wie die synaptische Kontrolle von Genexpression auf die plastischen Eigenschaften von Synapsen zurückwirkt.

    Leiter

    Michael R. Kreutz studierte Psychologie, Philosophie und Linguistik an der Universität Münster und promovierte anschließend in Verhaltens-Neurowissenschaften an der Ruhr-Universität Bochum. Anschließend wurde er wissenschaftlicher Mitarbeiter am Department of Brain and Cognitive Sciences am MIT, USA. Von 1990 bis 1993 war er wissenschaftlicher Mitarbeiter in der Abteilung Molekulare Neuroendokrinologie am Max-Planck-Institut für experimentelle Medizin in Göttingen.

    1993 zog er nach Magdeburg und leitet derzeit die Neuroplasticity Research Group (NPlast) am LIN.

    Seit Oktober 2015 hat er eine zweite Zugehörigkeit am Zentrum für Molekulare Neurobiologie (ZMNH) in Hamburg, wo er die Leibniz-Gruppe "Dendritische Organellen und synaptische Funktion" leitet.

    Projekte

    Im Zentrum der Arbeit unserer Forschergruppe steht die Frage, wie langfristige Veränderungen in der Struktur von Synapsen zu deren Rolle bei der Gedächtnisbildung beitragen. Eine Schlüsselrolle scheint hierbei die Kommunikation mit dem Zellkern zu spielen. 

    Wir untersuchen Mechanismen des Proteintransports aus Synapsen zum Zellkern, die Regulation von Genexpression und deren Bedeutung für die Stabilität und Funktion von Synapsen. Hier haben wir in den vergangenen Jahren wesentliche Fortschritte gemacht, auch wenn noch viele Fragen offenbleiben. Von großem Interesse für uns ist in diesem Zusammenhang vor allem, welche Relevanz die untersuchten Signalwege für die Alzheimer-Demenz haben.
     

    ProCognito - Pflanzliche Wirkstoffe zum Erhalt der kognitiven Leistungsfähigkeit im Alter

    Pflanzliche Wirkstoffe besitzen aufgrund ihrer evolutionären Entwicklung als Verteidigungssubstanzen ein besonderes Potential für neurologische Anwendungen und werden vom Menschen seit Jahr­tausenden genutzt. Als sessile Organismen verfügen Pflanzen weiterhin über viele antioxidative und antiinflammatorische Substanzen zur Stressbewältigung mit erwiesener positiver Wirkung am Menschen. In der Bevölkerung ist zudem eine hohe Akzeptanz zur Verwendung von natürlichen Produkten vorhanden. Im beantragten Projekt sollen daher die Kompetenzen der Verbundpartner PhytoAD und Metabolisches Syndrom aus der ersten Förderphase gekoppelt werden, um Wirkung und Anwendungspotential von ausgewählten Pflanzenextrakten und -stoffen spezifisch im Hinblick auf synaptische Prozesse zu untersuchen. Gleichzeitig stellt das IPB seine digitalisierte Substanzbibliothek, die derzeit die umfassendste Substanzsammlung mit natürlichen Wirkstoffen innerhalb Sachsen-Anhalts darstellt, im Rahmen des Forschungsverbundes 'Autonomie im Alter' für dieses Projekt, aber auch für weitere spezifische Fragestellungen und Anwendungen im Verbund zur Verfügung. Im Projekt gewonnene Daten werden in diese Datenbank eingepflegt und sind somit auch für zukünftige Anwendungen abrufbar. Um einen engen elektronischen Datenaustausch innerhalb des Verbundes zu ermöglichen, wird die unter Führung des IPB entwickelte Leibniz-Wirkstoff-Cloud (Leibniz-E-Center Bioactives) für den Forschungsverbund AiA geöffnet. Das Leibniz-E-Center Bioactives stellt die gemeinsam nutzbare Infrastruktur für die Suche, Analyse, Austausch sowie Management von Information und big data im Bereich Wirkstoffe und biologische Aktivität dar. Aufbau und Anwendung dieser Plattform soll von dem KMU Ontochem IT Solutions unterstützt werden, das patentierte Verfahren zur Implementierung von Ontologien, automatisiertem Text-Mining und Machine Learning zu medizinisch-pharmakologischen und Food-Anwendungen einbringt. Diese innovative elektronische Informations- und Kommunikationstechnologie-Anwendung soll auch über den Förderzeitraum hinaus einen nachhaltigen Mehrwert in der interdisziplinären Forschungskommunikation generieren.
